چکیده

The effect of exogenous auxin and cytokinin on cell proliferation and differentiation varies with the tissue system, and depends on the endogenous levels of the two hormones in the tissue. In this report, cytokinin (N-benzyladenine, BA) and auxin (indole-3-acetic acid, IAA) were used to treat young seedlings, leaf explants, and genetic tumors of a Nicotiana hybrid. Exogenous BA markedly accelerated genetic tumor formation in young seedlings, while exogenous IAA inhibited it on young seedlings but not on leaf explants. Genetic tumors showed low sensitivity to exogenous BA and IAA. RAPD analysis of the genomes of the genetic tumors and normal tissue suggested that tumor formation is related to genomic instability.

Response of various cucurbits to infection by plasmid-harboring strains of agrobacterium.

Tumor formation in cucurbit cultivars resulting from infection by various strains of Agrobacterium tumefaciens and Agrobacterium rhizogenes is environmentally affected. In all instances, tumors could be induced on excised cotyledons while inoculating attached cotyledons or stems resulted in no tumor formation. In addition, buttercup squash (Cucurbita maxima Duch. buttercup) was most susceptible...

TEACHING TOOLS IN PLANT BIOLOGYTM: LECTURE NOTES A Really Useful Pathogen, Agrobacterium tumefaciens

Bacteria of the genus Agrobacterium are very useful and unusual plant pathogens. Through a rare interkingdom DNA transfer, the bacteria move some of their genes into their host’s genome, thereby inducing the host cells to proliferate. The result is uncontrolled cell growth leading to a tumor (as induced by Agrobacterium tumefaciens and Agrobacterium vitis) or excessive production of roots (as i...

Agrobacterium rhizogenes GALLS protein substitutes for Agrobacterium tumefaciens single-stranded DNA-binding protein VirE2.

Agrobacterium tumefaciens and Agrobacterium rhizogenes transfer plasmid-encoded genes and virulence (Vir) proteins into plant cells. The transferred DNA (T-DNA) is stably inherited and expressed in plant cells, causing crown gall or hairy root disease. DNA transfer from A. tumefaciens into plant cells resembles plasmid conjugation; single-stranded DNA (ssDNA) is exported from the bacteria via a...

Attenuated Response to Agrobacterium Strains1

We have examined the response of the hormone-resistant mutants axrl and axr2 of Arabidopsis thaliana to inoculation by Agrobacterium tumefaciens and Agrobacterium rhizogenes. Our results indicate that recessive mutations in the axrl gene affect the frequency of tumor formation after inoculation with either Agrobacterium strain. In addition, tumors produced on axrl plants were smaller than those...

Relative Virulence of Agrobacterium Strains on Strawberry

Several strains of Agrobacterium tumefaciens and A. rhizogenes were shown to form tumors on runners of the diploid strawberry species Fragaria vesca L. Tumors, weighing from 0.1 to 8.3 mg, appeared from 2 to 4.5 weeks after infection. The majority of tumors tested for opine synthesis by high-voltage paper electrophoresis analysis showed positive results. These results demonstrate that diploid s...
